Avoiding Tech Transfer Disasters: A 6-Step Checklist for Small Pharma Teams

If you're a small pharma team (or even a solo project manager) about to hand off a drug candidate to a CDMO for the first time — this is for you. I've been in your shoes: managing tech transfer for a mid-stage asset, trying to coordinate data packages, and feeling like I was flying blind.

This checklist covers 6 steps I now run through before every tech transfer. It's built from my own mistakes (and the $30,000+ I've watched go down the drain because I skipped one).

Step 1: Lock Down Your Data Package Before Talking to Anyone

This sounds obvious. It is not obvious. Here's what I mean:

  • You need all batch records, analytical methods (including raw data), stability data, and deviation reports.
  • Make sure you have a data management plan for pharma clinical trials — a lot of small teams forget the batch-level raw data from early-stage CROs. I've seen vendors demand data in formats they can't read (like proprietary software outputs).
  • Checkpoint: Can you produce a complete data package in under 2 hours? If not, you're not ready. (Seriously: I spent 3 days digging through old emails for one transfer — never again.)

One mistake I made in 2021: I sent a PDF of analytical methods without the original instrument raw files. The CDMO rejected it. That cost us $2,000 in reanalysis and a 3-week delay. (Note to self: always include the .csv or .txt raw data.)

Step 2: Choose Your CDMO Based on Experience — Not Just Price

I have mixed feelings about price comparisons. On one hand, as a small team, every dollar matters. On the other hand, a cheap CDMO that doesn't understand your molecule will waste far more money than a slightly more expensive one that gets it right.

When I compared Vendor A (low bid, generic process) and Vendor B (specialist in your therapeutic area), side by side, the difference in success rate was staggering. Lonza is an obvious example of a CDMO with deep experience in both API manufacturing and cell culture media — but the point is to verify they have handled your specific type of molecule. Don't assume.

Checkpoint: Ask for at least 2 reference projects that match your drug's complexity. If they can't provide one, walk.

Step 3: Align on What 'Tech Transfer' Means — Put It in Writing

You'd think everyone uses the same definition. They don't. One CDMO I worked with thought tech transfer meant they'd receive a form and do everything else. Another thought it was a two-page summary. The disconnect cost us months.

According to ICH Q10 (Pharmaceutical Quality System), a good tech transfer includes: transfer of product knowledge, process knowledge, analytical methods, and related documentation. But that's high-level. You need to specify:

  • Who is responsible for each document?
  • What format does the data need to be in (e.g., CSV, XML, PDF)?
  • What is the timeline for review and approval?
  • How will deviations be handled?

(If I remember correctly, the FDA's 2019 guidance on tech transfer also recommends a detailed transfer plan — but don't quote me on the exact year. The principle stands.)

Step 4: Test the Data Flow with a Pilot Run

Don't wait until the actual transfer. Do a dry run: send a sample data package, ask them to confirm they can open and read everything, and then run a mock batch record. I started doing this after a nightmare in Q3 2023 where a vendor couldn't open our stability data files. The file format was standard (.xlsx), but their IT had blocked macros. Simple issue — but it shut us down for 4 days.

Checkpoint: Send a test dataset, wait for written confirmation that all files are accessible, and ask them to print a sample batch record on their template. That reveals formatting issues instantly.

Step 5: Map Out the Required SOPs and Regulatory Approvals Early

This is where small teams drop the ball. You might have an internal SOP for tech transfer, but the CDMO has their own. You need to reconcile them. Also: if your drug is for clinical trials, the transfer must comply with GMP (for Phase 2/3) or at least GMP-like standards. Lonza operates under GMP (their sodium hydroxide solution 0.5N is even marketed as GMP grade), so they know the drill — but you still need to align on what documents the FDA/EMA will expect.

I once got a rejection from the agency because the tech transfer report didn't include a certain stability data table. That table was sitting on our local drive, but it was never requested. So glad I now have a checklist that includes regulatory data requirements up front.

Checkpoint: Cross-reference your data package with the FDA Guidance for Industry: Technology Transfer (available on FDA.gov). If you can't check every box, flag it.

Step 6: Build in a 'Small Client Friendly' Escalation Path

When I was starting out, the vendors who treated my $500 orders seriously are the ones I still use for $50,000 orders. Tech transfer is similar: if a CDMO doesn't give you a dedicated contact person (even for small projects), run. You need someone who will escalate issues quickly, not a voicemail tree.

Checkpoint: Before signing, ask: "Who do I call on a weekend if the data transfer fails?" If the answer is vague, negotiate a better SLA or walk. There's no excuse for ignoring small clients — a good CDMO knows today's small biotech could be tomorrow's blockbuster.

Common Mistakes to Avoid

  • Assuming one format fits all: Some systems prefer CSV over XLSX; some prefer PDF/A over regular PDF. Test it.
  • Skipping the pilot run: I dodged a bullet once when the pilot run revealed that their data ingestion script crashed on our file naming convention. That was 10 minutes to fix. Without the pilot, it would have been a 3-day firefight.
  • Overlooking the tech transfer fee structure: Some CDMOs charge per document review. I had a $4,200 surprise bill because I didn't read the fine print. Ask upfront.
  • Not keeping your own copy of the final data package: Always keep an audit-trail version. You'll need it for the regulatory submission, not just the transfer.

Bottom line: tech transfer in pharma is as much about data management as it is about chemistry. If you treat it like a checklist rather than a mystery, you'll save time, money, and sanity. (And maybe, like me, you'll start documenting your own mistakes before they happen.)
