What I mean is that a supplier who lacks in-house solvent recycling will likely pass those costs and risks to you. I once had a vendor whose solvent recovery was outsourced; a logistical hiccup at their third-party facility delayed our delivery by a week. Now I ask upfront: “Do you recycle solvents on-site? What’s your typical recovery rate?” If they pause, I dig deeper.

Step 2: Assess Formulation Expertise (e.g., RPMI-1640 Compatibility)

If you’re sourcing APIs for cell culture media—say, RPMI-1640 formulations—you need a partner who understands the interaction between small molecules and the media matrix. I learned this the hard way. A supplier’s API looked fine on paper, but when we incorporated it into a DMEM batch, the pH shifted unexpectedly.

I now include a specific question in my RFQ: “Can you provide data on your API’s behavior in standard media formulations like RPMI-1640 or DMEM?” Companies like Lonza have built a reputation in this area precisely because they test across formulation contexts. Step 3: Scrutinize the GMP and Regulatory Track Record

  • Change control procedures—ask for a recent example of a process change and how it was communicated.

Per ICH Q3C guidelines, solvents are classified by risk, and a good CDMO will actively manage their Class 1 and 2 solvents. I reconcile this tension by framing the conversation around risk mitigation: “How do you ensure raw material availability for your top 10 products?” If they don’t have a clear answer, I move on.

Step 5: Run a Small Pilot Order (The Real Test)

Everything before this step is paper-based. The real test is a pilot order. I always start with a small, non-critical batch—maybe 10-20% of our typical volume. This reveals nuances no audit can capture:

  • How responsive are they during a real order, not just a sales call?
  • Do they provide COAs that match what they promised?
  • How do they handle minor issues (e.g., a slight delay in documentation)?

The most surprising insight from these pilots: vendor communication quality directly correlates with batch quality. The vendors who send proactive updates and clear, detailed invoices tend to deliver better products. It’s almost a quality perception halo effect—but the data backs it up in my experience.

Common Mistakes and Final Notes

Three mistakes I see repeatedly in our industry:

  1. Over-reliance on price per gram. The cheapest vendor often comes with hidden costs: re-testing, delayed timelines, or compliance remediation. The total cost of ownership—including your team’s management time—is the real metric.
  2. Assuming “small molecule” is a uniform category. There’s a world of difference between producing a simple salt and a complex, stereochemically demanding API. Match the vendor’s specialization to your molecule’s complexity.
